The ReadyPrep reduction-alkylation kit reduces and irreversibly blocks the formation of inter- and intramolecular disulfide bonds prior to isoelectric focusing (IEF). Disulfide bond formation is particularly problematic for basic proteins because of the increased rate of their formation in an alkaline environment. Moreover, many reducing agents become negatively charged during IEF and migrate off the IPG strip, thus allowing disulfide bonds to re-form (Görg 2000, Hoving 2002).
The ReadyPrep reduction-alkylation kit:
- Eliminates the potential for disulfide bond formation during IEF
- Reduces disulfide bonds with tributylphosphine (TBP) and alkylates the sulfhydryl groups with iodoacetamide (Herbert et al. 2001)
- Provides reagents for treatment of 100 samples (300 µl each, containing up to 10 mg/ml of protein)
- Performs treatment directly in the rehydration/sample buffer, in less than 2 hours, and with very little hands-on time

ReadyPrep™ Sequential Extraction Kit

The ReadyPrep sequential extraction kit is used to extract proteins of differing solubility from cell lysates. Based on the work of Molloy et al. (1998) and Herbert et al. (1998), the kit uses a series of three extraction buffers, each differing in their detergent and chaotrope concentrations (Rabilloud et al. 1997). The insoluble residues from each extraction step are further extracted with reagents of increasingly stronger solubilizing power.
The ReadyPrep sequential extraction kit:

ReadyPrep™ Protein Extraction Kit (Soluble/Insoluble)

The ReadyPrep protein extraction kit (soluble/insoluble) separates total protein samples into two fractions based on differential protein solubility. It generates a soluble fraction containing hydrophilic proteins and an insoluble fraction containing hydrophobic proteins. Proteins in the insoluble fraction are eventually resolubilized in ReadyPrep 2-D rehydration/solubilization buffer 1, a strongly chaotropic solution containing the zwitterionic detergent ASB-14. This simple fractionation strategy results in less complex samples and leads to improved identification of low-abundance proteins and a better overall view of the proteome.

ProteoMiner protein enrichment technology employs a large, highly diverse bead-based library of combinatorial peptide ligands, which simultaneously dilutes high-abundance proteins and concentrates low-abundance proteins. When a complex biological sample is applied to the beads, high-abundance proteins saturate their high-affinity ligand(s) and excess protein is washed away. In contrast, low-abundance proteins are concentrated on their specific affinity ligand(s).
ProteoMiner protein enrichment kits:

Bio-Rad’s mini cells resolve EcoRI or HindIII digests of lambda DNA in only 1.5 hours at 60 V. Small DNA fragments can be separated in as little as 15 minutes at 150 V.

Automation
The CHEF Mapper XA system offers two ways to optimize your separations. The unique auto-algorithm can automatically select optimal separation conditions, integrating 11 key variables and implementing starting separation conditions. Protocols can be refined using the Windows operating system-based interactive algorithm, which lets you specify several run variables simultaneously to derive optimal separation protocols.
Customization
Busy laboratories need equipment that can store and readily access key separation conditions. The CHEF Mapper XA system can store up to 99 programs.
Application Versatility
The CHEF Mapper XA system is the most flexible of the PFGE units. The CHEF Mapper XA system achieves higher resolution with greater speed and accuracy than any other PFGE system, making it ideal for all PFGE applications. The CHEF Mapper XA system lets you choose any pulse angle from 0 to 360°, which allows optimal separation of both chromosomal and plasmid DNA with one system. Accurate sizing of fragments requires an expanded linear range of separation. Switch-time ramps increase the mobility of fragments by gradually changing the switch times during the course of a run. Nonlinear (for example, concave or convex) ramps change the switch-time increments from the start to the end of a run. These nonlinear ramps separate fragments linearly from 50 to 700 kb, yielding more precise fragment size measurements. The multistate mode of the CHEF Mapper XA system enhances resolution in selected fragment size ranges. Each vector (angle of pulse) can be assigned its own voltage (field intensity) and its own switch time (duration of pulse). Up to eight different states can be combined into one run to optimize the separation of subsets of fragments in the sample. The application of secondary pulses of defined voltage, duration, angle, and frequency can enhance the separation and resolution of very large DNA molecules by releasing DNA caught in the gel matrix.

Chelex 100 chelates polyvalent metal ions, with a selectivity for divalent over monovalent ions of approximately 5,000 to 1. The resin avidly binds divalent cations such as Mg2+, inactivating metallo-nucleases and other enzymes, as well as binding other polyvalent metal ions that interfere with enzyme-based applications such as PCR and ligation. Due to the high selectivity for divalent over monovalent ions, Chelex 100 Molecular Biology Grade Resin can be used for DNA and RNA preparations from samples with high levels of salts.
• Chelex 100 eliminates time-consuming and labor-intensive steps required with conventional nucleic acid extraction methods. A typical DNA or RNA extraction protocol involves: add Chelex 100 suspension to the sample, boil to release nucleic acids and denature sample, spin to pellet resin beads and sample debris, use supernatant for downstream steps such as PCR or RT-PCR.

MicroRotofor lysis kits provide cell lysis and protein extraction protocols that are tailored to the specific needs of different sample sources. Convenient and easy to use, all four kits are based on the same chaotropic protein solubilization buffer (PSB), which contains nondetergent sulfobetaine 201 (NDSB 201) along with urea, thiourea, and CHAPS for particularly effective solubilization (Vuillard et al. 1995). The kits generate total protein samples that are ready for fractionation with the MicroRotofor liquid-phase IEF cell, but they may also be applied to other downstream applications, such as SDS-PAGE and 2-D gel electrophoresis.
Species-Specific Protocols
Different sample types have different requirements for effective cell disruption, and all four kits combine PSB with other elements to accommodate these specific needs.
- MicroRotofor cell lysis kit (mammal) — uses maceration and solubilization (up to a 100 mg sample) using ReadyPrep mini grinders and PSB (Vuillard et al. 1995). The kit is intended for use with samples from mammalian sources, but it may also be applied to other animal tissues
- MicroRotofor cell lysis kit (bacteria) — for use with both gram-negative and gram-positive bacterial cultures. It employs enzymatic digestion using lysozyme followed by solubilization into PSB (Vuillard et al. 1995)
- MicroRotofor cell lysis kit (plant) — for use with soft plant tissues and cultured cells. It employs maceration (up to a 1 g sample) and solubilization into PSB. A ReadyPrep 2-D cleanup kit is provided to enable removal of plant-derived phenolics, which may interfere with IEF
- MicroRotofor cell lysis kit (yeast) — for use with yeast cultures (~60 µl wet cell pellet). It employs enzymatic digestion of the cell wall with lyticase followed by solubilization into PSB (Scott et al. 1980, Vuillard et al. 1995)

Droplet Digital PCR Technology and Workflow
The QX200 Droplet Digital PCR System consists of two instruments, the QX200 Droplet Generator and the QX200 Droplet Reader, and their associated consumables.
The QX200 Droplet Generator is used to partition ddPCR reaction mix into thousands of nanoliter-sized droplets. After PCR on a thermal cycler, droplets from each sample are analyzed individually on the QX200 Droplet Reader. Droplets are read well by well as they are streamed single file past a two-color optical detection system in a serial manner. Up to 96 samples can be processed per run. The PCR-positive and PCR-negative droplets are counted to provide absolute quantification of target DNA in digital form. Alternatively, amplified products can be extracted from droplets following PCR for downstream applications, such as sequencing or cloning.
Additional Instruments
The QX200 Droplet Digital PCR System requires the use of the C1000 Touch Thermal Cycler with 96–Deep Well Reaction Module and the PX1 PCR Plate Sealer.

The ChemiDoc XRS+ System optimizes reproducibility and reliability of experimental data, enabling quantitative comparisons between different experiments. Using proprietary algorithms, this imaging system is calibrated at setup to ensure:

- Appropriate flat fielding correction is automatically and consistently applied to image data for every application

The ChemiDoc XRS+ System offers fast and sensitive chemiluminescence detection in addition to gel and blot documentation of fluorescent and colorimetric samples. The system includes a supersensitive 16-bit CCD camera that is supercooled for detection of faint samples. Bio-Plex 3D Instrument
The system comes with all instrument components combined in one shell, a PC, an arm-mounted monitor, keyboard, mouse, sheath fluid, an empty sheath container, a calibration kit and performance verification kit (shipped separately), and an accessory kit with probes, heater blocks, and cables. The system comes with the base version of the xPONENT 4.0 software. As an added value, the system will also come with Bio-Plex Manager software version 6.0 for data analysis.

21CFR Part 11 Module
xPONENT software provides users the opportunity to enable the 21CFR Part 11 module for regulatory compliance. This module maintains data integrity through the use of electronic signatures. The 21CFR Part 11 module contains the Basic Security module.
LIS Module
This module enables xPONENT 4.0 software to transmit and receive data from laboratory information systems (LIS) and laboratory information management systems (LIMS) using an ASTM file format.
Automation Module
The xPONENT 4.0 software facilitates communication/integration between front-end automation devices and Bio-Plex 3D system. The front-end automation provider can build the required driver through an application programming interface (API), which will be supplied with the automation interface module.
Extra Seat Licenses
Extra software seats allow software to be installed on PCs for data analysis away from the instrument. The system comes with one instrument and two desktop PC licenses. Extra seats come in groups of 3. The maximum number of seat licenses per system is 15.
Swivel Base
The system can be purchased with an optional swivel base that allows the unit to rotate by 90° in either direction. This can simplify connection of this system to certain automation platforms. The swivel base can be purchased either at the same time as the instrument or later.
